爆破强度性能分析:ASTM F2392是一种比较软组织上丁基防水嵌缝胶的爆裂或破裂强度的方法。将新鲜猪肺剪成大小均一,直径为4cm的圆片,在中间打一个直径为3 mm的小孔,放入生理盐水中备用。分别将丁基防水嵌缝胶液和交联剂溶液加入AB胶混合注射器的两侧,等体积混合注射到肺的表面并堵住孔洞,密封胶厚度控制在1 mm25 ℃等待成胶。将堵漏后的肺组织安装在爆破强度测试仪上,仪器下方通入生理盐水记录破裂时的最大值(最大抗爆破压力)。测试后的肺组织及其表面丁基防水嵌缝胶用福尔马林溶液固定2d,包埋切片并用苏木精一伊红(HE)染色,使用数字切片扫描系统(生产公司:匈牙利3DHISTECH公司,产品型号)扫描并进行截面观察。体外降解性能测定:将丁基防水嵌缝胶浸泡在超纯水中脱盐2d,然后冻干称量(mo)。冻干样品在PBS缓冲液(pH=7.4中浸泡24 h以上,以备后续实验使用。将复溶胀的丁基防水嵌缝胶完全浸入胶原酶浓度为200g/mL的PBS溶液中,并在37 ℃200 r/min的条件下振荡2 h4 h6 h8 h10 h和12h。到点将溶液离心并移除液体,清洗沉淀物,冻干称量,记作m,。质量残留率(W)根据式(4)计算细胞毒性测定:在%孔板中接种1X10"个/孔的L-929细胞悬液,细胞预培养24 h。24 h后换液,向培养板各孔中加入浸泡了24 h的丁基防水嵌缝胶浸提液,放入培养箱中孵育24 h。向每孔中加入10 L CCK-8溶液,放入培养箱中孵育2h。用酶标仪测定450中伯胺官能团的含量,通过比较伯胺官能团的减少比例得知,C10-CFG的疏水基团C10实际取代度为5.4%。如Fig.2(a)所示,CAT-CFG与CAT-CIO-CFG溶液在278 nm波长处的紫外吸收峰相较CFG溶液增大,该吸收峰与CAT单体的吸收峰重叠,证明成功修饰上了儿茶酚基团CAT。在Fig.2(b)中,测定CFGCAT-CFG及CAT-C 10-CFG溶液在278 nm波长处的吸光度后代入标准曲线,求得单位质量中CAT基团的含量,再和TNBS法定量的未改性丁基防水嵌缝胶中伯胺官能团的总量比较,得知CAT-CFG和CAT-C 10-CFG的CAT基团取代度分别为1.6%和1.2%。丁基防水嵌缝胶性能评价丁基防水嵌缝胶成胶时间:根据可看出,3种改陛丁基防水嵌缝胶成胶时间(约5 e皆短于未改性丁基防水嵌缝胶约8e说明C10基团的疏水性作用和CAT基团的氧化反应可促进丁基防水嵌缝胶迅速固化。其中双重改性CAT-C10-CFG成胶时间与C10-CFG和CAT-CFG相比无明显差异,说明疏水作用和CAT氧化反应不会互相干扰。Fig.3 (a)中的交联剂浓度为0.15 g/mL,而Fig.3(b)中的交联剂浓度为0.2 g/mL,与丁基防水嵌缝胶液等体积混合后,丁基防水嵌缝胶中的伯胺基团与交联剂中NHS活性基团之比分别为3:3和3:4,交联剂浓度的提升并未缩短成胶时间,这可能是因为伯胺基团与 NHS基团之比为3:3时己达到饱和交联的状态。http://www.sdyuantai.com/
